【摘要】:Therangeofconcentrationofafluorescingsubstanceinsolutionoverwhichthefluorescencevarieslinearlywiththeconcentrationistherangemostusefulforquantitativeanalysis.Thisrangeisaffectedbypropertiesofthesolutionunderanalysisandbyfeaturesofthemeasuringsystem.Thistestmethodprovidesameansoftestingtheperformanceofafluorescencemeasuringsystemandofdeterminingtheconcentrationrangeoverwhichthesystemissuitableformakingagivenquantitativeanalysis.Thistestmethodisnotmeantforcomparingtheperformanceofdifferentfluorescencemeasuringinstruments.1.1Thistestmethodcoversaprocedureforevaluatingthelimitsofthelinearityofresponsewithfluorescenceintensityoffluorescence-measuringsystemsunderoperatingconditions.Particularattentionisgiventoslitwidths,filters,andsamplecontainers.Thistestmethodcanbeusedtotesttheoveralllinearityunderawidevarietyofinstrumentalandsamplingconditions.Theresultsobtainedapplyonlytothetestedcombinationofslitwidthandfilters,andthesize,typeandilluminationofthesamplecuvette,allofwhichmustbestatedinthereport.Thesourcesofnonlinearitymaybethemeasuringelectronics,excessiveabsorptionofeithertheexcitingoremittedradiation,orboth,andthesamplehandlingtechnique,particularlyatlowconcentrations.1.2Thistestmethodhasbeenappliedtofluorescence-measuringsystemsutilizingcontinuousandlow-energyexcitationsources(forexample,anexcitationsourceof450-Welectricalinputorless).Thereisnoassurancethatextremelyintenseilluminationwillnotcausephotodecompositionofthecompoundssuggestedinthistestmethod.Forthisreasonitisrecommendedthatthistestmethodnotbeindiscriminatelyemployedwithhigh-intensitylightsources.Itisnotatestmethodtodeterminethelinearityofresponseofothermaterials.Ifthistestmethodisextendedtoemployotherchemicalsubstances,theprincipleswithincanbeapplied,butnewmaterialparameters,suchastheconcentrationrangeoflinearity,mustbeestablished.Theusershouldbeawareofthepossibilitythattheseothersubstancesmayundergodecomposition,oradsorptionontocontainers.1.3Thistestmethodhasbeenappliedtofluorescence-measuringsystemsutilizingasingledetector,thatis,aphotomultipliertubeorasinglephotodiode.Ithasnotbeendemonstratedifthismethodiseffectiveforphoto-arrayinstrumentssuchasthoseusingaCCDoradiodearraydetector.1.4Thistestmethodisapplicableto10-mmpathlengthcuvetteformatsandinstrumentscoveringawavelengthrangewithin190to900nm.Theuseofothersampleformatshasnotbeenestablishedwiththistestmethod.Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
